Custom Sample Placement for Photosynthesis & Respiration Assays
Step 1: Experiment Setup
Instructions:
1. Select your experiment type above
2. Click on wells in the 96-well plate below to assign samples
3. Assign at least one BLANK and 3-4 pH STANDARDS for calibration
4. Upload CSV files from your plate reader for the wavelengths shown
5. Generate and select the best calibration curve
Step 2: Sample Placement
Sample Requirements:
• Blank: 200µL of 0.01M bicarbonate buffer (no indicator) - Required for background subtraction
• Standards: 200µL total volume at known pH values
• Photosynthesis Assay: Uses thymol blue & cresol red indicator mix (pH range 6.6-9.2)
• Respiration Assay: Uses bromothymol blue & bromocresol purple indicator mix (pH range 6.0-8.2)
• Duplicates Recommended: Using duplicate standards improves accuracy, but single standards are acceptable if conserving materials

Step 3: Upload Absorbance Data
Upload the CSV file exported from your Accuris MR9600 plate reader at the required wavelength for your assay (595 nm for Photosynthesis, 450 nm for Respiration). The tool will automatically extract absorbance values for your selected wells.
Step 4: Select Best Calibration Approach
Select the calibration method with the highest R² value. The tool tests multiple wavelength combinations to find the best fit.
Step 5: Review & Save Calibration
Import Calibration Profile
Import a calibration JSON file created on another device (e.g., teacher's calibration shared with students).
